Overview

The interferon alpha (IFN-α) signaling pathway is a critical component of the innate immune response, primarily involved in antiviral defense. IFN-α is a type I interferon produced by various cell types upon detection of viral infection. Once secreted, IFN-α initiates a complex intracellular signaling cascade that leads to the expression of hundreds of interferon-stimulated genes (ISGs), which mediate antiviral, immunomodulatory, and antiproliferative effects. The pathway involves binding to the IFNAR receptor, activation of JAK kinases, phosphorylation of STAT proteins, formation of the ISGF3 complex, and transcriptional activation of ISGs. Dysregulation can contribute to autoimmune diseases or impaired immunity; therapeutic modulation is used in some cancers and chronic viral infections.
