The interferon gamma (IFN-γ) pathway is a critical signaling cascade in the immune system, primarily involved in inflammation, cell-mediated immunity, and host defense against pathogens and tumors. IFN-γ is secreted mainly by activated T helper 1 (Th1) cells, cytotoxic T lymphocytes (CTLs), natural killer (NK) cells, macrophages, mucosal epithelial cells, and B-cells. The pathway involves the binding of IFN-γ to its receptor, activation of JAK kinases, phosphorylation and dimerization of STAT1, translocation of STAT1 to the nucleus, and subsequent regulation of gene transcription. It plays a crucial role in various biological functions, including macrophage activation, MHC expression, and immune regulation. Dysregulation of the pathway can contribute to autoimmune diseases, chronic inflammatory states, and cancer.
