Overview

Interferon-gamma receptor is a heterodimeric cell-surface receptor complex composed of two chains, IFNGR1 and IFNGR2, which together form the high-affinity receptor for the cytokine interferon-gamma (IFN-γ), the sole type II interferon in humans[1][2][3][4]. Upon IFN-γ binding, IFNGR1 and IFNGR2 associate and initiate intracellular signaling via the JAK1 and JAK2 kinases, leading to phosphorylation and activation of STAT1 and downstream transcription of interferon-stimulated genes central to host defense, immune modulation, and cell growth[1][4][5][6]. Mutations in either subunit cause severe immunodeficiency, especially with recurrent or severe mycobacterial infections[6][7]. As a critical node in immune signaling, IFNGR is an important therapeutic target in autoimmunity, infection, and immuno-oncology[6][7][8].
