Overview

Interferon-induced protein 35 (IFI35) is an interferon-stimulated protein upregulated in response to type I and II interferons. It interacts with proteins such as N-Myc-interacting protein (Nmi) and casein kinase 2-interacting protein-1 (CKIP-1) to regulate its stability and function. IFI35 modulates multiple cellular processes, acting as both a positive and negative regulator of viral replication, depending on context and viral strain. It is expressed in a variety of cell types and can translocate from the cytoplasm to the nucleus upon stimulation. Besides its intracellular signaling roles, IFI35 can function extracellularly as a damage-associated molecular pattern to promote inflammation. Disease associations include cancers, viral infections, and inflammatory disorders. To date, IFI35 remains a promising immunomodulatory target, although no direct therapeutics are approved

Mechanism of action

Modulation of IFI35 would impact *type I interferon signaling* and *NF-kappaB pathways*. IFI35 interacts with and can drive degradation of RIG-I via K48-linked ubiquitination, affecting antiviral signaling. In the context of viral infection, interference with IFI35-NS1 interactions could be exploited for antiviral drug development, as in the case of swine influenza.
