Overview

The interferon receptor family consists of a group of structurally related class II helical cytokine receptors, each forming a heterodimeric or homodimeric complex at the cell surface to transmit signals from the three major interferon types. Type I interferon receptors (comprising IFNAR1 and IFNAR2) mediate responses to interferon-alpha, beta, and related cytokines, leading to broad antiviral and immunomodulatory effects. Type II interferon receptor (IFNGR1 and IFNGR2) transmits the effects of interferon-gamma, central to cellular immune responses and macrophage activation. Type III interferon receptor (IFNLR1 and IL10RB) specializes in mucosal antiviral defense and signals similarly to type I but with restricted tissue distribution. All signal through the JAK/STAT pathway to regulate genes for immune defense, cell growth, and apoptosis. Therapeutically, their agonists and antagonists have applications in infectious diseases, cancer, autoimmunity, and inflammatory disorders, but the broad downstream effects require careful management due to safety concerns stemming from immune activation
