Overview

Interferon-related developmental regulator 2 (IFRD2) is a protein broadly expressed in human tissues and is a member of the interferon-related developmental regulator family. IFRD2 acts as a negative regulator of mRNA translation by directly binding to ribosomes, inserting into the mRNA exit channel, and physically obstructing mRNA passage. This results in repression of translation and engagement of the ribosome in a noncanonical manner. Structurally, IFRD2 contains a core Armadillo-type fold with several HEAT repeats, and a C-terminal helix that interacts deeply with ribosomal RNA and proteins. Although initially described as a transcriptional regulator, current cryo-EM studies provide strong evidence for its primary function as a translational repressor. IFRD2 is highly expressed in tissues involved in development and differentiation but is not associated with any known therapeutically targeted disease mechanism or drug interaction.
