Overview

Lymphokines are a functional subset of cytokines produced by lymphocytes, particularly T-helper cells, that act as chemical messengers to coordinate the immune response [1][3]. They play a critical role in cell-mediated immunity by regulating the growth, differentiation, and activation of various immune cells, including macrophages, B cells, and other T cells [2][8]. In modern pharmacology, the term is increasingly superseded by the more specific 'cytokine' or 'interleukin' nomenclature, as lymphokines encompass a diverse array of proteins such as Interleukin-2 (IL-2) and Interferon-gamma [13][15]. Dysregulation of lymphokine production is central to the pathogenesis of autoimmune diseases, chronic inflammation, and certain malignancies like renal cell carcinoma and melanoma [10][21]. Therapeutic strategies involving lymphokines include the use of recombinant versions to boost immune activity against cancer (e.g., Aldesleukin) or the development of monoclonal antibodies to inhibit specific pro-inflammatory lymphokines [16][19]. However, systemic administration of lymphokines or their targeted activation of immune cells is often limited by significant therapeutic challenges, such as vascular leak syndrome or cytokine release syndrome [16][22].
