Overview

Interleukin 1 receptor accessory protein-like 2 (IL1RAPL2) is a member of the interleukin 1 receptor family that shares similarity with accessory proteins in this group, especially IL1RAPL1[1][2][5]. It is encoded by the IL1RAPL2 gene, which is found on the X chromosome, and it is highly expressed in the brain, suggesting potential involvement in neuronal signaling and cognitive functions[2][3]. The protein is structurally characterized by multiple immunoglobulin domains and participates in protein-protein interactions at synapses, though its precise ligand and downstream signaling functions remain under investigation[2]. Genetic variants in the IL1RAPL2 locus have been associated with X-linked non-syndromic cognitive disability, implicating this protein in neurological development and function[1][2]. While implicated in rare diseases such as Muckle-Wells syndrome and Adult-Onset Still’s Disease, there are currently no known approved drugs directly targeting IL1RAPL2[2].
