Interleukin-1 receptor antagonist (IL-1Ra) (IL-1Ra)

Interleukin-1 receptor antagonist (IL-1Ra) is a naturally occurring protein that serves as a critical endogenous regulator of the inflammatory response. It belongs to the interleukin-1 family and functions by binding with high affinity to the interleukin-1 receptor type I (IL-1RI) without inducing a biological signal, thereby competitively blocking the pro-inflammatory actions of IL-1α and IL-1β (Arend, 2002, Cytokine & Growth Factor Reviews). This inhibition is vital for maintaining immune homeostasis; an imbalance between IL-1 and IL-1Ra is a hallmark of various autoinflammatory and autoimmune conditions, including rheumatoid arthritis and the rare genetic disorder Deficiency of IL-1 Receptor Antagonist (DIRA) (Aksentijevich et al., 2009, NEJM).
