Interleukin-1 receptor-associated kinase 1 (IRAK-1) is a key serine/threonine kinase that functions as a critical mediator in the innate immune response and inflammatory signaling pathways [1, 8]. It is a member of the IRAK family and plays a central role in signal transduction downstream of Toll-like receptors (TLRs) and Interleukin-1 receptors (IL-1Rs) [3, 7]. Upon receptor activation, IRAK-1 is recruited to the myddosome complex, where it is phosphorylated by IRAK-4, leading to the activation of downstream transcription factors such as NF-kappaB and AP-1 [2, 12]. Dysregulation or overexpression of IRAK-1 is associated with various pathological conditions, including autoimmune diseases, chronic inflammatory disorders, and several types of cancer, such as acute myeloid leukemia (AML) and myelodysplastic syndromes (MDS) [1, 10, 11]. Consequently, IRAK-1 has emerged as a significant therapeutic target, with small molecule inhibitors like pacritinib demonstrating clinical efficacy by blocking its kinase activity and reducing the production of pro-inflammatory cytokines [3, 5, 13]. Recent therapeutic strategies also include the development of selective IRAK-1 degraders and covalent inhibitors to overcome challenges related to scaffolding functions and drug resistance [1, 4].
