IRAK3 is a member of the interleukin-1 receptor-associated kinase family, primarily expressed in myeloid cells. It functions as a negative regulator of innate immune signaling pathways downstream of Toll-like receptors (TLRs) and interleukin-1 receptors (IL-1R). Unlike other IRAK family members, IRAK3 is considered a pseudokinase and inhibits TLR and IL-1R signaling by stabilizing the Myddosome complex, preventing activation of downstream kinases. It is involved in tumor progression, asthma, and sepsis.
