IRAK4 is a serine/threonine protein kinase that plays a central role in the innate immune response, particularly in signaling pathways downstream of Toll-like receptors (TLRs) and interleukin-1 receptors (IL-1Rs). It acts as a master regulator at the top of TLR/IL-1R signaling cascades. Upon receptor activation, IRAK4 autophosphorylates and activates itself, subsequently phosphorylating downstream targets such as IRAK1. This leads to activation of transcription factors like NF-kappaB and AP-1, resulting in production of inflammatory cytokines. It is a therapeutic target for autoimmune diseases, inflammation-driven cancers, and other inflammatory conditions.
