Interleukin-1 receptor-like 2 (IL1RL2) is a cell surface cytokine receptor of the interleukin 1 receptor family, most commonly referred to as the IL-36 receptor. Upon binding IL-36 agonists (IL-36α, IL-36β, IL-36γ), it forms a signaling complex with IL-1 receptor accessory protein (IL-1RAcP) and activates NF-κB and MAPK intracellular pathways, resulting in robust inflammatory cytokine and chemokine production. IL1RL2 is expressed especially in epithelial (e.g., skin keratinocytes) and immune cell types such as dendritic cells, playing a pivotal role in amplifying local and systemic immune responses. Dysregulated IL1RL2 signaling contributes to several skin and autoimmune diseases, most notably various forms of psoriasis, arthritis, and inflammatory bowel disease; antagonists of this pathway (e.g., anti-IL-36R antibodies or IL-36RA) are under investigation for controlling disease-associated inflammation and tissue damage[1][3][4].
