Overview

Interleukin-1 Receptor-like protein (SIGIRR), also known as TIR8 or IL-1R8, is a negative regulator of inflammation belonging to the interleukin-1 receptor/toll-like receptor superfamily. It inhibits signal transduction from multiple interleukin and toll-like receptors, suppressing downstream signaling pathways such as NF-κB and JNK. SIGIRR plays a crucial role in maintaining immune homeostasis and acts as a tumor suppressor. Its downregulation is associated with increased susceptibility to inflammation-driven diseases and cancer progression, making it a potential therapeutic target for autoimmune diseases, chronic inflammatory conditions, and certain cancers.
