Interleukin-10-producing dendritic cells (DC-10) are a specialized subset of human tolerogenic dendritic cells (tDCs) that are essential for the induction of Type 1 regulatory T (Tr1) cells (Gregori et al., 2010, Blood). They are characterized by the expression of CD14, CD16, CD141, and CD163, along with high levels of the tolerogenic molecule HLA-G and the cytokine IL-10 (Amodio et al., 2015, Frontiers in Immunology). Biologically, DC-10 cells maintain immune homeostasis by suppressing effector T cell responses and promoting peripheral tolerance (Comi et al., 2020, Frontiers in Immunology). In therapeutic settings, DC-10 are not traditional molecular targets but are instead used as a cellular product or induced ex vivo using agents like IL-10, Vitamin D3, and dexamethasone to treat autoimmune diseases and prevent transplant rejection (Gregori & Roncarolo, 2018, Frontiers in Immunology). Their clinical application focuses on their ability to restore tolerance in conditions like Crohn's disease and Graft-versus-Host Disease (GvHD).
