The Interleukin 10 receptor beta subunit (IL10RB) is a shared cell surface receptor required for activation of five class II cytokines: IL-10, IL-22, IL-26, IL-28 (IFNλ2), and IFNL1/3. It acts as an accessory chain that is essential for forming functional heterodimeric or heterotetrameric cytokine receptors with various alpha subunits. Upon ligand binding, it initiates intracellular signaling via the JAK/STAT pathway, ultimately modulating gene transcription. Defects in IL10RB are associated with early-onset inflammatory bowel disease and other immunological disorders.
