The Interleukin-12 receptor (IL-12R) is a type I cytokine receptor that mediates the effects of Interleukin-12 (IL-12), a heterodimeric cytokine essential for host defense. The IL-12 receptor consists of two subunits: IL-12Rβ1 and IL-12Rβ2. It is primarily expressed on T cells and natural killer (NK) cells, where it promotes the differentiation of naive T cells into Th1 cells, enhances the cytotoxic activity of NK cells and CD8+ T cells, and stimulates the production of interferon-gamma (IFN-γ) and tumor necrosis factor-alpha (TNF-α). IL-12 also exhibits anti-angiogenic activity. Signal transduction involves the recruitment of Janus family tyrosine kinase 2 (JAK2) and signal transducer and activator of transcription 4 (STAT4). IL-12 has been explored as a potential therapeutic agent in cancer and autoimmune diseases.
