Overview

The interleukin-12 receptor beta subunit refers to two distinct but related proteins, IL-12 receptor subunit beta 1 (IL12RB1) and IL-12 receptor subunit beta 2 (IL12RB2), which are critical components of the interleukin-12 (IL-12) receptor complex. These type I transmembrane proteins belonging to the hemopoietin receptor superfamily form a high-affinity binding site for IL-12, enabling downstream signaling essential for immune responses. The functional receptor is a heterodimer of IL-12Rβ1 and IL-12Rβ2. IL-12Rβ1 binds to the p40 subunit shared by both IL-12 and IL-23. IL-12Rβ2 is primarily responsible for signal transduction, coupling with the JAK2/STAT4 pathway upon ligand binding. Deficiency or mutations in either subunit can result in severe immunodeficiencies.
