Overview

The interleukin-13 receptor (IL-13R) is a type I cytokine receptor that primarily binds interleukin-13 (IL-13), a key cytokine involved in immune regulation, particularly in allergic inflammation and asthma. The functional IL-13 receptor complex is formed by the association of two subunits: IL-13Rα1 and the shared alpha chain, IL-4Rα. Signaling proceeds through activation of Janus kinases (JAKs), leading to phosphorylation and dimerization of STAT proteins—primarily STAT6—which then translocate into the nucleus to regulate target gene expression involved in Th2 immune responses such as allergy and asthma. Therapeutically targeting this pathway has been effective in treating severe allergic diseases due to its pivotal role in driving pathological inflammation.
