Overview

Interleukin-13 (IL-13) signaling via the IL-4 receptor alpha (IL-4Rα) primarily occurs through the type II receptor complex, composed of IL-4Rα and IL-13Rα1. IL-13 binds to IL-13Rα1, which then recruits IL-4Rα to form the functional receptor. This complex activates downstream signaling pathways, notably the JAK/STAT6 pathway, leading to gene expression changes associated with Th2 responses, alternative macrophage activation, and the promotion of allergic inflammation, airway hyperresponsiveness, and fibrosis. IL-13Rα2 acts as a decoy receptor, attenuating IL-13 signaling.
