Overview
Interleukin-15 (IL-15) is a pleiotropic cytokine belonging to the 4-alpha helix bundle family that plays a central role in both innate and adaptive immunity. It modulates the development, survival, proliferation, and activation of various immune cell types, including Natural Killer (NK) cells, CD8+ memory T cells, NKT cells, γδ T cells and ILC1 cells. Dysregulated IL-15 production has been implicated in autoimmune diseases and cancer. Recombinant forms and agonists are under development for cancer immunotherapy.

Mechanism of action
IL-15 interacts with a heterotrimeric receptor complex consisting of IL-15Rα, IL-2/IL-15Rβ, and the common γ chain (*γc*), activating downstream signaling pathways like JAK1/JAK3 → STAT3/STAT5, p38 MAPK, and NF-kB to promote lymphocyte proliferation, survival, and activation.
