Interleukin 17 (IL-17) modulators are molecules—often small molecules or biologics—that influence the activity of IL-17 cytokines, particularly IL-17A. These modulators can either inhibit or enhance the signaling pathways mediated by IL-17, which is a key pro-inflammatory cytokine involved in immune responses and various inflammatory and autoimmune diseases. IL-17 plays a central role in innate immunity, inflammation, and osteoclastogenesis. Excessive or inappropriate IL-17 activation contributes to autoimmune/inflammatory diseases (psoriasis, rheumatoid arthritis, psoriatic arthritis, asthma, periodontitis), cancer progression, and bone pathology. Therapeutic modulation involves small molecule inhibitors/modulators targeting the interaction between IL‑17A and its receptor, and biologic drugs targeting either the cytokine itself or its receptor.
