Overview

Act1, also known as NF-κB activator 1, is a crucial adaptor protein in the IL-17 signaling pathway. It mediates immune and inflammatory responses by connecting IL-17 receptor activation to downstream cascades, particularly NF-κB. Act1 possesses an N-terminal helix-loop-helix domain and a C-terminal coiled-coil region. It is expressed in various cell types, including B cells, epithelial cells, and monocytes. Its primary function is to act as an adaptor, bridging IL-17 receptors to effectors. Upon IL-17 binding, Act1 interacts with the receptor and activates pathways like NF-κB and JNK/SAPK. Recent evidence also suggests a direct role in nuclear transcriptional regulation. Because it is central to the inflammatory signaling—especially Th17-driven processes—Act1 is being investigated as a potential therapeutic target for autoimmune diseases.
