Overview

The Interleukin 17 signaling pathway is mediated by IL-17 family cytokines (primarily IL-17A and IL-17F) binding to multimeric cytokine receptor complexes composed of different IL-17 receptor subunits, most frequently IL-17RA paired with IL-17RC. Upon activation, these receptors initiate intracellular signaling via adaptors such as Act1, activating downstream mediators including NF-κB, MAPK, and others, resulting in the induction of inflammatory genes, chemokine production, antimicrobial peptide expression, and tissue remodeling. Dysregulation of IL-17 signaling is implicated in autoimmune and inflammatory diseases, as well as certain cancers and infections, making these receptors important drug targets.
