Overview

Interleukin-17D is a secreted cytokine that belongs to the IL-17 family, primarily expressed in non-hematopoietic tissues such as muscle, brain, adipose tissue, heart, lung, and pancreas. IL-17D can stimulate endothelial cells to produce pro-inflammatory mediators including IL-6, IL-8 (via an NF-κB–dependent pathway), and GM-CSF, while inhibiting hematopoiesis of myeloid progenitor cells. IL-17D also plays an immunomodulatory role in cancer by recruiting natural killer (NK) cells, promoting an anti-tumor microenvironment, and suppressing metastasis. In infectious disease models, loss of IL-17D is associated with improved pathogen clearance and enhanced CD8+ T cell function, suggesting that it may suppress adaptive immune responses during infection by regulating dendritic cell activity. There is no current evidence that IL-17D itself is directly targeted by drugs or acts as a biomarker for patient selection, but its roles in cancer immunology and infection highlight its complexity as a therapeutic target[2][4].
