Overview

Interleukin 2-inducible T-cell kinase (ITK) is a non-receptor tyrosine kinase belonging to the Tec family. ITK plays a crucial role in T cell signaling, activation, and differentiation, particularly in Th2, Th9, and Th17 cell effector functions. It is involved in the regulation of cytokine production and is essential for adaptive immunity. ITK is activated downstream of the T cell receptor (TCR) and phosphorylates key substrates like PLCγ1 and LAT/LCP2, leading to calcium flux, NFAT activation, and downstream signaling cascades involved in lymphokine production, proliferation, and differentiation. ITK is a therapeutic target for autoimmune diseases, allergic disorders, and certain cancers.
