Overview

The interleukin-2 receptor beta and gamma subunits (IL-2Rβ/CD122 and IL-2Rγ/CD132, the common gamma chain) are integral membrane proteins that together form a key part of the type I cytokine receptor family. They assemble with the IL-2 receptor alpha chain to constitute the high-affinity IL-2 receptor complex on activated T cells, while the beta and gamma subunits alone can form an intermediate-affinity dimeric receptor, present on memory T and natural killer (NK) cells. IL-2 binding triggers receptor heterodimerization and signal transduction through associated Janus kinases (primarily JAK1 and JAK3), leading to STAT phosphorylation, nuclear translocation, and gene activation critical for lymphocyte proliferation, survival, and immune function. The gamma chain is shared with several other cytokine receptors (IL-4, IL-7, IL-9, IL-15, IL-21), and its genetic loss results in severe immune deficiency (X-linked SCID). Pharmacological targeting of the IL-2 receptor pathway modulates immune responses for treatment of autoimmunity, cancer, organ transplantation, and immune deficiencies.
