Overview

The **interleukin-2 receptor beta and gamma chains** (IL-2Rβ/γ) are two essential subunits of the heterotrimeric interleukin-2 receptor complex found primarily on lymphocytes. Together, the β (CD122) and γ (CD132 or common gamma chain) subunits form a receptor that mediates the biological effects of the cytokine interleukin-2 (IL-2), supporting the proliferation, survival, and differentiation of T cells and natural killer (NK) cells. The gamma chain is shared among several cytokine receptors (including those for IL-4, IL-7, IL-9, IL-15, and IL-21), while the beta chain is more specific to IL-2 and IL-15 receptor signaling. The IL-2Rβ/γ chains are central to immune homeostasis and are involved in both immune activation and tolerance. Mutations in the gamma chain cause X-linked severe combined immunodeficiency (X-SCID), highlighting their critical role in lymphoid development and function. Drugs targeting this receptor complex are used in immunosuppressive regimens for transplantation and under investigation or in use for cancer immunotherapy.
