Overview

The Interleukin-2 receptor subunit gamma (IL2RG)-mediated common gamma-chain cytokine receptor complex is a group of type I cytokine receptors that share the IL2RG subunit, also known as the common gamma chain (γc) or CD132 (UniProt P31785). This shared subunit is essential for the formation of functional receptor complexes for interleukin-2 (IL-2), IL-4, IL-7, IL-9, IL-15, and IL-21, which are critical regulators of immune system development and homeostasis (PubMed: 25624443). These complexes signal primarily through the Janus kinase 3 (JAK3) and Signal Transducer and Activator of Transcription 5 (STAT5) pathways to promote the proliferation and differentiation of T cells, B cells, and natural killer (NK) cells (PubMed: 11244038). Mutations in the IL2RG gene lead to X-linked severe combined immunodeficiency (X-SCID), a life-threatening condition characterized by the absence of functional T and NK cells (NIH: GARD). In clinical practice, these complexes are targeted by immunosuppressive drugs like basiliximab and daclizumab to prevent transplant rejection, while JAK inhibitors like tofacitinib are used to treat autoimmune conditions by blocking downstream signaling (DrugBank: DB00075, DB00848). Furthermore, therapies utilizing IL-2 or IL-15 agonists aim to stimulate these receptor complexes to enhance anti-tumor immunity in various cancers (PubMed: 28438888).
