The Interleukin-22 receptor (IL-22R) is a type II cytokine receptor that mediates the biological effects of interleukin-22 (IL-22). The functional IL-22 receptor complex is a heterodimer composed of IL-22R1 and IL-10R2. Upon ligand engagement, the receptor complex activates JAK/STAT and other signaling pathways, leading to transcriptional activation and diverse biological responses including epithelial barrier maintenance, antimicrobial peptide production, tissue repair, and inflammation modulation. Dysregulation of IL-22R signaling is implicated in autoimmune diseases.
