Interleukin-24 (IL-24) receptor complexes are heterodimeric transmembrane proteins belonging to the Type II cytokine receptor family [1, 2]. IL-24 signals through two distinct receptor combinations: the IL-20R1/IL-20R2 complex (Type I IL-20 receptor) and the IL-22R1/IL-20R2 complex (Type II IL-20 receptor) [2, 4]. Upon binding, these receptors primarily activate the Janus kinase/signal transducer and activator of transcription (JAK/STAT) signaling pathway, specifically STAT1 and STAT3, to regulate gene expression [2, 3]. While IL-24 is uniquely recognized for its ability to selectively induce apoptosis in a wide range of cancer cells without harming normal cells, its receptors are also expressed in non-transformed tissues where they mediate inflammatory responses, particularly in the skin and joints [3, 4]. Consequently, these receptor complexes are investigated as therapeutic targets for oncology via gene therapy or recombinant protein agonism, and for autoimmune conditions such as psoriasis and rheumatoid arthritis via receptor antagonism [4].
