The Interleukin-25 receptor (IL-25R) is a heterodimeric receptor complex composed of IL17RA and IL17RB. It serves as the functional receptor for interleukin 25 (IL-25) and interleukin 17B (IL-17B), playing a central role in mediating type 2 immune responses. Activation of IL-25R promotes the secretion of type 2 cytokines, enhances humoral immunity, and recruits/activates eosinophils, basophils, and mast cells. Due to its role in orchestrating type 2 immune responses, IL-25R is a potential therapeutic target for allergic diseases and certain infections, while its overactivation can contribute to chronic inflammatory diseases like asthma or atopic dermatitis.
Agonist binding initiates JAK-STAT, p38 MAPK, and NF-κB signaling pathways, leading to the production of type 2 cytokines (IL-4, IL-5, IL-9, IL-13) and subsequent immune responses.
