Interleukin 27 (IL-27) is a heterodimeric cytokine belonging to the IL-12 family, composed of IL-27p28 and EBI3 subunits. Produced primarily by activated antigen-presenting cells, IL-27 binds to a receptor complex of IL-27Rα and gp130, activating JAK-STAT signaling. IL-27 exhibits pleiotropic effects on T cell subsets, promoting Th1 and Tr1 differentiation while inhibiting Th2 and Th17. It enhances cytotoxicity in CD8+ T cells and supports germinal center reactions in B cells. Displaying both pro-inflammatory (IFNγ production) and anti-inflammatory (IL-10 induction) properties, therapeutic targeting of IL-27 is under investigation for infectious diseases, autoimmunity, and cancer.
