Interleukin-28 receptor alpha (IL28RA, also called IFN-lambda receptor 1 or IFNLR1) is a transmembrane protein that forms the high-affinity component of the type III interferon receptor complex. It pairs with interleukin-10 receptor beta (IL10RB) to form a functional receptor for type III interferons (interleukin-28A, interleukin-28B, interleukin-29—collectively known as IFN-lambdas)[1][2][3]. Upon ligand binding, IL28RA initiates signaling primarily through the JAK/STAT pathway, leading to the induction of interferon-stimulated genes that confer antiviral and immunomodulatory effects[3]. Expression is highest in epithelial tissues; its signaling is generally less inflammatory and more localized compared to type I interferon receptors, which makes it an attractive target for antiviral and immunotherapeutic development[3].
Drugs or biologicals targeting IL28RA would modulate the JAK/STAT signaling pathway by interfering with or mimicking type III interferon signaling[3]. Indirect manipulation via administration of type III interferons enhances antiviral gene expression with limited pro-inflammatory signaling[3].
