Overview

Interleukin 31 (IL-31) is a cytokine primarily produced by activated CD4+ T lymphocytes, especially T helper 2 (TH2) cells. It signals through a heterodimeric receptor complex composed of IL-31 receptor A (IL-31RA) and Oncostatin M receptor beta (OSMRβ), activating JAK/STAT, PI3K/AKT, and MAPK pathways. IL-31 plays a key role in immune modulation, neuroimmune communication (inducing itch), and tissue effects in the skin, lungs, and intestines. It is clinically significant in diseases like atopic dermatitis and prurigo nodularis, and therapeutic targeting of IL-31 or its receptor has shown promise in reducing pruritus.
