Overview

Interleukin 36 Receptor (IL36R) is a member of the interleukin-1 receptor family and forms a heterodimeric receptor complex with IL-1RAcP. Activation by its agonists (IL-36α, IL-36β, IL-36γ) triggers pro-inflammatory signaling pathways, while antagonists (IL-36Ra, Interleukin-38) block receptor activation. IL36R is predominantly expressed in epithelial tissues and plays a key role in innate and adaptive immunity. Dysregulation of IL36R signaling is implicated in various inflammatory diseases, making it a therapeutic target for conditions such as psoriasis and inflammatory bowel disease.

Mechanism of action

Agonist binding induces heterodimerization with IL-1RAcP, activating downstream NF-kB and MAPK signaling pathways. Antagonists block IL-1RAcP recruitment, preventing signal transduction.
