Overview

The Interleukin-4 receptor (IL-4R) is a type I cytokine receptor complex that plays a critical role in type II immune responses and allergic inflammation. It exists in two forms: Type I (IL-4Rα + γc), which binds only IL-4, and Type II (IL-4Rα + IL-13Rα1), which binds both IL-4 and IL-13. Upon ligand binding, the receptor activates JAK kinases and STAT6, leading to gene expression changes involved in Th2 differentiation and IgE class switching. The soluble form of IL-4R acts as a decoy receptor, modulating immune response intensity. IL-4R is a therapeutic target for various allergic and inflammatory diseases.
