Overview

The interleukin 5 receptor alpha subunit (IL-5Rα) is a receptor for interleukin-5 (IL-5), a cytokine critical for the development, activation, survival, and function of eosinophils. It forms a heterodimeric receptor complex with the common beta chain (βc), which is shared with the receptors for interleukin-3 (IL-3) and granulocyte-macrophage colony-stimulating factor (GM-CSF). IL-5Rα is primarily expressed on eosinophils, basophils, and some B cells. Binding of IL-5 to IL-5Rα triggers intracellular signaling cascades, primarily via the JAK/STAT pathway, leading to the biological effects of IL-5. As a key regulator of eosinophil biology, IL-5Rα is a therapeutic target for eosinophil-mediated diseases, such as asthma and hypereosinophilic syndromes. Monoclonal antibodies and peptide antagonists targeting IL-5Rα are being developed to block IL-5 signaling and reduce eosinophil activity.
