Overview

The Interleukin 5 receptor beta chain (IL-5Rβ), also known as CD131 or the common beta chain (βc), is a shared subunit of the IL-3, IL-5, and GM-CSF receptors. It does not bind IL-5 directly, but forms a high-affinity receptor complex with the IL-5 specific alpha chain (IL-5Rα) upon ligand binding. The beta chain is crucial for signal transduction, activating JAK kinases and downstream signaling pathways involved in eosinophil proliferation and differentiation. As a therapeutic target, blocking IL-5 signaling through the beta chain is of interest for treating eosinophil-driven diseases like asthma and allergic inflammation. Because it is shared among multiple cytokine receptors (IL‑3R & GM-CSFR), it plays broader roles in hematopoiesis regulation.
