Overview

Interleukin 6 antisense RNA 1 (IL6-AS1) is a long noncoding RNA (lncRNA) that is transcribed antisense to the IL6 gene locus. Unlike canonical therapeutic targets such as receptors or enzymes, IL6-AS1 does not encode a protein but regulates the expression of interleukin 6 (IL-6) at both transcriptional and post-transcriptional levels[2][4]. In the cytoplasm, IL6-AS1 acts as an endogenous sponge, binding to microRNA miR-149-5p and stabilizing IL-6 mRNA, leading to increased IL-6 production. In the nucleus, IL6-AS1 recruits early B-cell factor 1 (EBF1) to the IL-6 promoter, promoting histone modifications (H3K4 methylation, H3K27 acetylation) to enhance IL-6 transcription. IL6-AS1 is upregulated in chronic obstructive pulmonary disease and is linked to airway inflammation through multiple mechanisms that augment IL-6 levels[2][4]. There is currently no evidence that IL6-AS1 directly interacts with pharmacological agents or serves as a therapeutic target, but its function as a regulator of cytokine expression suggests emerging translational interest.
