The Interleukin-6 (IL-6) receptor family is a group of cytokine receptors that mediate the biological effects of the IL-6 family of cytokines. These receptors are characterized by their use of a common signal-transducing subunit, gp130 (IL6ST), which is essential for downstream signaling. Family members include IL6R, IL11R, CNTFR, LIFR, OSMR, IL27RA, and IL31RA. Signaling is mediated through the JAK/STAT, Ras/Raf/MAPK, and PI3K/PKB pathways. The IL-6 family regulates diverse physiological processes including hepatic acute phase response, B-cell stimulation and differentiation, balance between regulatory and effector T cells, metabolic regulation, and neural functions. Blockade or modulation of this family's signaling has therapeutic potential in autoimmune diseases.
