The interleukin-7 receptor alpha subunit (IL-7Rα), also known as CD127, is a type I cytokine receptor. It serves as a critical component of two distinct heterodimeric receptors: the interleukin-7 receptor (with the common gamma chain, γc/CD132) and the thymic stromal lymphopoietin (TSLP) receptor (with TSLPR). It is expressed primarily on lymphoid lineage cells and plays a crucial role in lymphocyte development, survival, and homeostasis. Altered expression/function is implicated in various diseases, including immunodeficiencies, autoimmunity, and cancer, making it a potential therapeutic target.
Targeted therapeutics may exploit this axis for cancer immunotherapy. Soluble form can modulate immune responses by acting as a decoy or regulator in circulation.
