Overview

Interleukin-7 receptor subunit alpha (IL-7Rα), also known as CD127, is a type I cytokine receptor essential for the development and homeostasis of T and B lymphocytes. It forms a heterodimer with the common gamma chain (γc/CD132) to create the functional IL-7 receptor. Upon binding of IL-7, the receptor complex activates intracellular signaling pathways, primarily the JAK/STAT pathway, leading to cell survival and proliferation. IL-7Rα also participates in the TSLP receptor complex and is implicated in various diseases, including SCID, autoimmune disorders, and leukemia.

Mechanism of action

Ligand binding (IL-7 or TSLP) to the IL-7Rα/γc heterodimer activates the JAK/STAT signaling pathway, promoting cell survival and proliferation. Receptor endocytosis regulates signaling duration and receptor availability.
