Internal jugular vein stenosis is a clinical condition, not a molecule or classical therapeutic target. It refers to the abnormal narrowing of the internal jugular vein (IJV), which is a major venous outflow tract for the brain. Stenosis can be caused by intraluminal factors (e.g., congenital membranes, webs, thrombi) or extrinsic compression (most commonly by elongated styloid process, C1 vertebra hypertrophy, neck masses, or vascular abnormalities)[1][5]. This can result in impaired venous drainage from the brain and is associated with symptoms such as headache, dizziness, tinnitus, insomnia, neck pain, and cognitive disturbances[3][5][6]. Associations between IJVS and conditions like idiopathic intracranial hypertension, multiple sclerosis, Ménière disease, and Alzheimer’s disease have been described, though causality is debated[2][5]. Diagnosis is by CT or MR venography and management may be conservative, endovascular (e.g., vein stenting), or surgical decompression in select cases[1][5][6]. There is significant anatomical variability in jugular vein size, and normal narrowing is common at the skull base and upper cervical spine, adding diagnostic complexity[4]. Note: IJVS is not a discrete protein, receptor, transporter, or gene product. It is a vascular pathology, so it does not fulfill criteria for a canonical therapeutic target.
