Intestinal barrier epithelial cells comprise a monolayer of specialized cell types—**enterocytes**, **goblet cells**, **Paneth cells**, **tuft cells**, and microfold cells—forming the gut lining that separates the lumen from underlying tissue. Their principal role is to control nutrient absorption and fluid transport, protect against pathogens and toxins through the barrier and mucosal defense mechanisms, and regulate local immune responses in concert with immune and stromal cells. These cells are rapidly renewed from stem cells in the crypts (turnover every 2-7 days)[2][4]. Tight junctions and other intercellular adhesion structures are critical for maintaining barrier integrity[1][3][5]. Disruption of this barrier is implicated in inflammatory bowel diseases, colorectal cancer, and infection[1].
