Overview

Intestinal epithelial cell proliferation is the process by which new cells are generated within the lining of the intestine. This highly dynamic and tightly regulated process is essential for maintaining intestinal homeostasis, supporting nutrient absorption, and providing a barrier against pathogens. The proliferative compartment is located at the base of crypts where intestinal stem cells divide to produce transient amplifying progenitor cells which then differentiate into mature epithelial lineages. Proliferation is controlled by Wnt/β-catenin signaling, EGF signaling, TGF-alpha/beta peptides, nutrient sensing, and microbiome influence. Dysregulation can lead to insufficient renewal (barrier dysfunction) or excessive proliferation (neoplasia).
