Overview

Intestinal epithelial cell receptors (IECRs) encompass a heterogeneous population of proteins expressed on the apical and basolateral membranes of the intestinal mucosa. These receptors are fundamental to gastrointestinal physiology, mediating the absorption of essential nutrients, the secretion of ions and water, and the sensing of both dietary components and the gut microbiota (NIH, 2021). Key subclasses include G protein-coupled receptors (GPCRs) involved in metabolic signaling, Toll-like receptors (TLRs) that initiate innate immune responses, and specialized transporters like the neonatal Fc receptor (FcRn) (PubMed, 2020). In clinical practice, these receptors are critical in the pathogenesis of inflammatory bowel disease (IBD) and celiac disease, where barrier dysfunction and aberrant signaling lead to chronic inflammation. Therapeutic strategies often involve modulating specific IECRs, such as the guanylate cyclase-C receptor targeted by linaclotide for irritable bowel syndrome, or targeting the downstream pathways of immune-sensing receptors to restore mucosal homeostasis (StatPearls, 2023).
