Overview

Intestinal epithelial cell tight junction proteins are a group of transmembrane and cytoplasmic adaptor proteins that form specialized intercellular contacts at the apical region between adjacent intestinal epithelial cells. These complexes serve two main functions: they act as a selective barrier regulating paracellular transport (“gate” function) and maintain cellular polarity by separating apical from basolateral membrane domains (“fence” function). The principal transmembrane components include occludin, various claudin isoforms, and junctional adhesion molecules (JAMs). These interact with cytoplasmic scaffold proteins such as ZO family members that link them to the actomyosin cytoskeleton. Disruption or dysregulation of these tight junctions is implicated in diseases characterized by increased intestinal permeability—such as inflammatory bowel disease, infections, and certain cancers—by allowing luminal antigens, pathogens, or toxins to cross into underlying tissue. While individual tight junction components may be studied as potential therapeutic targets or biomarkers for gut barrier dysfunction, “intestinal epithelial cell tight junction proteins” refers collectively to this multi-protein structural assembly rather than a single molecular entity.
