Overview

The intestinal epithelial tight junction protein complex is a group of structural and signaling proteins that form the primary physical barrier between intestinal epithelial cells. This complex includes transmembrane proteins such as occludin, claudins, and junctional adhesion molecules (JAMs), as well as intracellular scaffold proteins like zonula occludens 1, 2, and 3. These complexes regulate the selective permeability of the intestinal epithelium, preventing pathogenic bacteria and harmful substances from entering underlying tissues while allowing nutrient absorption. Disruption or dysfunction of these complexes leads to increased intestinal permeability ("leaky gut"), which is implicated in various diseases including IBD, IBS, celiac disease, infections by enteric pathogens (*E. coli*, *Salmonella*, etc.), and other inflammatory conditions. While not a single molecular target for drug development in the traditional sense (such as a receptor or enzyme), modulation of tight junction integrity is an area of therapeutic interest for restoring barrier function in gastrointestinal diseases.
