Overview

Intestinal epithelial tight junction proteins form specialized intercellular junctions located at the apical part of the epithelium between neighboring cells. They constitute a critical component of the intestinal barrier, regulating paracellular permeability and maintaining epithelial integrity. These dynamic protein complexes consist of transmembrane proteins (occludin, claudins, JAMs, tricellulin) that interact across the intercellular space and peripheral proteins (ZO-1, ZO-2, ZO-3) that connect to the cytoskeleton. Their proper function is essential for intestinal health, and their dysregulation is implicated in various pathological conditions including inflammatory bowel disease, obesity, and other metabolic disorders.
